C-erbB-2和Her-2人類基因是因其與逆轉錄病毒V—erbB-2相似而被發(fā)現(xiàn)。之后Coussens L及Schechte AL等對Neu,Her-2,C-erbB-2進行測序及染色體定位分析,證明他們是同一基因。
人類該基因定位于染色體17q12,其編碼產(chǎn)物為185kD的跨膜精蛋白p185,由1255個氨基酸組成,720—987位屬于酪氨酸激酶區(qū)。Her-2/neu蛋白是具有酪氨酸蛋白激酶活性跨膜糖蛋白,是EGFR家族成員之一。
Her-2/neu蛋白由胞外的配體結合區(qū)、單鏈跨膜區(qū)及胞內(nèi)的蛋白酪氨酸激酶區(qū)三部分組成,由于目前尚未發(fā)現(xiàn)能與Her-2/neu蛋白直接結合的配體.其主要通過與家族中其他成員包括EGFR(HERl/erbBI),HER3/erbB3。HER4/erbB4形成異二聚體而與各自的配體結合。Her-2/neu蛋白常為異二聚體*伴侶,且活性常強于其它異二聚體。當與配體結合后,主要通過引起受體二聚化及胞漿內(nèi)酪氨酸激酶區(qū)的自身磷酸化.激活酪氨酸激酶的活性。

The human genes of C-erbB-2 and Her-2 are discovered because they are similar to retrovirus V - erbB-2. After Coussens L and Schechte AL, Neu, Her-2, C-erbB-2 were sequenced and chromosomal location analysis proved that they were the same gene.
The human gene is located on chromosome 17q12, and its encoded product is 185kD transmembrane protamine p185, which is composed of 1255 amino acids. The 720 to 987 belongs to the tyrosine kinase region. Her-2/neu protein is a tyrosine protein kinase active transmembrane glycoprotein and is one of the members of the EGFR family.
Her-2/neu protein by extracellular ligand binding region, a single transmembrane region and intracellular protein tyrosine kinase domain is composed of three parts, because has not yet found the ligand can bind directly to Her-2/neu protein. It mainly through other members with family including EGFR (HERl/erbBI), HER3/ erbB3. HER4/erbB4 forms an isooligomer and combines with the respective ligands. The Her-2/neu protein is often the preferred mate of the isomer, and the activity is often stronger than the other iso - two - polymer. When binding to the ligand, the activity of tyrosine kinase is activated mainly by causing the receptor dimerization and the autophosphorylation of the tyrosine kinase region in the cytoplasm.
It has been shown that overexpression of HRE-2 can potentially activate EGFR signaling pathways, and also promote EGFR mediated transformation and tumor formation.
