一般培養(yǎng)基用1．05kg /cm2，121．3℃15—30分鐘可達到*滅菌的目的。滅菌的溫度及維持的時間隨滅菌物品的性質(zhì)和容量等具體情況而有所改變。例如含糖培養(yǎng)基用0．56kg/cm2（8磅/英寸2），112．6℃滅菌15分鐘，但為了保證效果，可將其他成分先行121．3℃，20分鐘滅菌，然后以無菌操作手續(xù)加入滅菌的糖溶液。又如盛于試管內(nèi)的培養(yǎng)基以1．05kg/cm2，121．3℃滅菌20分鐘即可，而盛于大瓶內(nèi)的培養(yǎng)基以1．05kg/cm2滅菌30分鐘。
蒸汽壓力所用單位為kg/cm2（千克/厘米2），它與lb/in2（磅/英寸2）和溫度的換算關(guān)系見表Ⅵ-5。
 實驗室中常用的高壓蒸汽滅菌鍋有立式、臥式和手提式等幾種，其構(gòu)造如圖Ⅵ-4。本實驗介紹手提式高壓蒸汽滅菌鍋的使用方法。

實驗原理
