4、免疫組化中抗原修復(fù)的*條件是什么？（尤其是微波修復(fù)）
關(guān)于抗原修復(fù)，我試過的修復(fù)條件有EDTA熱修復(fù)，檸檬酸鈉為緩沖液的微波修復(fù)以及高壓鍋修復(fù)。從我的實驗結(jié)果來看，微波修復(fù)其實很不容易掉片子的，而EDTA熱修復(fù)和高壓鍋修復(fù)是很容易掉片子的。因為掉片子主要是因為加熱過程中產(chǎn)生的氣泡所引起，而微波修復(fù)水加熱到沸騰，沾在片子上的氣泡就會少，不會因為小氣泡上串引起脫片。做EDTA修復(fù)時，你可以將片子靠的盡量近些，或是在載玻片四周墊些棉花什么的，然后蓋上蓋玻片，這樣修復(fù)的話就能夠盡量減少載玻片上小氣泡的形成。我們用的微波修復(fù)條件為：2.94 g檸檬酸三鈉溶于1000 ml的水，調(diào)PH值至6.0，微波修復(fù)10分鐘。你可以試試，時間可以根據(jù)需要自己調(diào)整。對于檸檬酸修復(fù)來說，只要高壓修復(fù)時間控制在加閥冒氣后90秒，冷水下終止開高壓鍋蓋，高壓修復(fù)和微波修復(fù)一樣不掉片子，而且一般高壓修復(fù)效果往往更好一點；關(guān)于EDTA熱修復(fù)，一般說明書上都講的是PH值等于9，實際上我試過，如果單傳用EDTA，很容易掉片子，但是用Tris-EDTA,一般就不會掉片子了，Tris-base的濃度為0.05 mmol/L,高壓和微波修復(fù)都可以，PH也等于9。
