8、加一抗
將溫箱中的載玻片取出，用吸水紙擦干載玻片反面和正面組織周圍的血清，加一抗，如果做對(duì)照實(shí)驗(yàn)，就在對(duì)照的組織上加PBS。加完一抗后于4 oC冰箱中保存過(guò)夜。
9、加二抗
將載玻片從冰箱中取出，放入PBS中洗3次，每次5 min，擦干組織周圍的PBS后加上二抗,然后置于37 oC溫箱中半小時(shí)。
10、加SABC
將片子從溫箱中取出, 放入PBS中洗3次，每次5 min，擦干組織周圍的PBS后加上SABC， 然后置于37 oC 溫箱中半小時(shí)。SABC稀釋100倍（990 ul PBS：10 ul SABC）。
11、加顯色劑
將片子從溫箱中取出, 放入PBS中洗3次，每次5 min，擦干組織周圍的PBS后加上顯色劑。（顯色劑的配置：在1 ml水中加1滴顯色劑A，搖勻，然后加1滴顯色劑B，搖勻，再加1滴顯色劑C，搖勻）   A：DAB      B：H2O2    C：磷酸緩沖液
12、復(fù)染
將顯色后的片子用清水沖洗一段時(shí)間后，浸泡于蘇木精中染色，一般動(dòng)物組織為半分鐘，植物組織3-5 min。

8, plus a resistance
Remove the slides from the incubator, wipe the sera around the opposite side of the slide and the frontal tissue with blotting paper, and add a primary antibody. If done as a control, add PBS to the control tissue. After the addition of a primary antibody in a 4 oC refrigerator overnight.
9, plus two anti
Remove the slide from the freezer and wash in PBS for 3 times for 5 min. Wipe off the PBS around the tissue and add the secondary antibody. Place the slide in a 37 ° C incubator for half an hour.
10, plus SABC
Remove the pellet from the incubator, wash it in PBS for 3 times for 5 min, dry the tissue around the PBS, add SABC, and place in a 37 ° C incubator for half an hour. SABC was diluted 100-fold (990 ul PBS: 10 ul SABC).
11, plus color agent
Remove the tablets from the incubator and wash them in PBS for 3 times for 5 minutes. Dry the PBS around the tissue and add the color reagent. (Developer configuration: add 1 drop of developer A in 1 ml of water, shake well, then add 1 drop of developer B, shake well, add 1 drop of developer C, shake well) A: DAB B : H2O2 C: phosphate buffer
12, counterstain
The color after the film rinse with water for a period of time, soaked in hematoxylin staining, the general animal tissue for half a minute, plant tissue 3-5 min.
