檢驗原理
用抗原包被微量板孔,制成固相載體。加患者血清到板孔中,其所含的抗體特異性地與固相載體中現(xiàn)存抗原結(jié)合,形成免疫復(fù)合物。除去多余物質(zhì)后,加入結(jié)合了堿性磷酸酶的IgG、IgA或IgM抗體,使之與上述免疫復(fù)合物反應(yīng)。洗板,除去多余的結(jié)合物,加入底物(對硝基苯磷酸鹽)。其與酶結(jié)合的免疫復(fù)合物反應(yīng),產(chǎn)生有顏色產(chǎn)物,顏色強度與特異性抗體含量成正比。

霍亂弧菌能還原硝酸鹽為亞硝酸鹽,靛基質(zhì)反應(yīng)陽性,當培養(yǎng)在
含硝酸鹽及細菌氨酸的培養(yǎng)基中,產(chǎn)生靛基質(zhì)與亞硝酸鹽,在濃
硫酸存在時,生成紅細菌,稱為霍亂紅反應(yīng),但其他非致病性弧
菌亦有此反應(yīng),故不能憑此鑒定霍亂弧菌。EL Tor型霍亂弧菌與
古典型霍亂弧菌生化反應(yīng)有所不同。前者Vp陽性而后者為陰性。
前者能產(chǎn)生強烈的溶血素,溶解羊紅細胞,在血平板上生長的菌
落周圍出現(xiàn)明顯的透明溶血環(huán),古典型霍亂弧菌則不溶解羊紅細
胞。個別EL Tor型霍亂弧菌株亦不溶血。
抗原性
根據(jù)弧菌O抗原不同,分成Ⅵ個血清群,第Ⅰ群包括霍亂弧菌的兩
個生物型。第Ⅰ群A、B、C三種抗原成份可將霍亂弧菌分為三個血
清型:含AC者為原型(又稱稻葉型),含AB者為異型(又稱小川
型),A、B、C均有者稱中間型(彥島型)。
抵抗力
霍亂弧菌古典生物型對外環(huán)境抵抗力較弱,EL-Tor生物型抵抗力
較強,在河水、井水、海水中可存活1~3周,在鮮魚,貝殼類食
物上存活1~2周。
霍亂弧菌對熱,干燥,日光,化學(xué)消毒劑和酸均很敏感,耐低溫
,耐堿。濕熱55℃,15分鐘,100℃,1~2分鐘,水中加0.5ppm
氯15分鐘可被殺死。0.1%高錳酸鉀浸泡蔬菜、水果可達到消毒目
的。在正常胃酸中僅生存4分鐘。能發(fā)酵葡萄糖、麥芽糖、甘露醇
、蔗糖、半乳糖等產(chǎn)酸產(chǎn)氣,遲緩發(fā)酵乳糖。不分解阿拉伯糖、
水楊素、衛(wèi)矛醇、鼠李糖、木糖、側(cè)金盞花醇和肌醇。氧化酶、
明膠液化試驗陽性。V-P試驗大多數(shù)EL-Tor生物型陽性,而古典
生物型大多為陰性?;魜y弧菌能分解細菌氨酸產(chǎn)生吲哚,同時能
還原硝酸鹽為亞硝酸鹽。因此當霍亂弧菌培養(yǎng)在含硝酸鹽的蛋白
胨水中,所產(chǎn)生的亞硝酸鹽與吲哚結(jié)合成亞硝基吲哚,滴加濃硫
酸即出現(xiàn)薔薇細菌,稱為霍亂紅試驗陽性。
