6.膽汁七葉苷水解試驗
(1)原理:在10%~40%膽汁存在下,測定細菌水解七葉苷的能力。七葉苷被細菌分解生成的七葉素,七葉素與培養(yǎng)基中的枸櫞酸鐵的二價鐵離子發(fā)生反應形成黑色化合物。
(2)方法:將被檢菌接種于膽汁七葉苷培養(yǎng)基中,35℃孵育18~24h后,觀察結果。
(3)結果:培養(yǎng)基*變黑為陽性,不變黑為陰性。
(4)應用:主要用于鑒別D群鏈球菌與其它鏈球菌的區(qū)別,以及腸桿菌科的某些種、某些厭氧菌(如脆弱擬桿菌等)的初步鑒別。D群鏈球菌本試驗為陽性。
7.淀粉水解試驗
(1)原理:產(chǎn)生淀粉酶的細菌能將淀粉水解為糖類,在培養(yǎng)基上滴加碘液時,可在菌落周圍出現(xiàn)透明區(qū)。
(2)方法:將被檢菌劃線接種于淀粉瓊脂平板或試管中,35℃孵育18~24h,加入革蘭碘液數(shù)滴,立即觀察結果。
(3)結果:陽性反應,菌落周圍有無色透明區(qū),其它地方藍色;陰性反應,培養(yǎng)基全部為藍色。
(4)應用:用于白喉棒狀桿菌生物型的分型,重型淀粉水解試驗陽性,輕、中型陰性;芽胞桿菌屬菌種和厭氧菌某些種的鑒定。
8.甘油復紅試驗
(1)原理:甘油可被細菌分解生成丙酮酸,丙酮酸脫去羧基為乙醛,乙醛與無色的復紅生成醌式化合物,呈深紫紅色。
(2)方法:取被檢菌接種于甘油復紅肉湯培養(yǎng)基中,于35℃孵育,觀察2~8d。應同時做陰性對照。
(3)結果:紫紅色為陽性,與對照管顏色相同為陰性。
(4)應用:主要用于沙門菌屬內(nèi)各菌種間的鑒別。傷寒沙門菌、甲(丙)型副傷寒沙門菌、豬霍亂沙門菌、孔道夫沙門菌和仙臺沙門菌本試驗為陰性,乙型副傷寒沙門菌結果不定,其它不常見沙門菌多數(shù)為陽性。
4. β-galactosidase test (ONPG test)
(1) Principle: lactose permease and β-galactosidase can rapidly decompose in the process of lactose fermentation. Some bacteria only galactosidase, which can only ferment lactose slowly, all the rapid fermentation of lactose and slow fermentation of bacteria can be rapid hydrolysis of o-nitrophenyl-β-D-galactopyranoside (O-nitrophenyl-β-D-galactopyranoside , ONPG) to generate yellow o-nitrophenol. (2) Method: Inoculated bacteria to be tested in ONPG broth, incubated at 35 ℃ water bath or incubator for 18 ~ 24h, the results were observed.
(3) Results: showing bright yellow is positive, colorless is negative.
(4) Application: It can be used for the rapid identification of lactic acid bacteria which are slowly fermenting. The bacteria in this method can be positive for bacteria rapidly and slowly decomposing lactose. Escherichia, Citrobacter, Klebsiella, Hafnia, Serratia and Enterobacter are positive tests, and Salmonella, Proteus and Providencia Staphylococcus is negative.
5. VP test
(1) Principle: The ability of bacteria to produce methyl acemethanol was determined. Some bacteria, such as Enterobacter aerogenes, decompose glucose to pyruvate, and pyruvate decarboxylates to form acetylmethylmethanol. Acetylmethylmethanol is oxidized to diacetyl under basic conditions, which in turn combines with guanidine-containing substances such as arginine in the medium to form red compounds. V-P test is positive.
(2) Method: Inoculate the bacteria to be inoculated in glucose peptone water and incubate at 35 ℃ for 24-48h. Add 50ml of α-naphthol (95% ethanol solution) 50ml, shake the tube gently and add 0.2ml 400g / L KOH, gently shake the tube 30s to 1min, and then stand the observation results.
(3) the results: red were positive, yellow or copper-like negative.
(4) Application: Mainly used for the identification of Escherichia coli and Enterobacter aerogenes. This test is often used in conjunction with the MR test, under normal circumstances, the former is a positive bacteria, the latter is often negative, and vice versa. However, the Enterobacteriaceae bacteria do not necessarily have such a law, such as honeycomb Haemophilus and Proteus VP test and MR test are often positive.
