【預(yù)期用途】 豬乙型腦炎又名日本乙型腦炎是由豬乙腦病毒（Porcine Japanese Encephalitis Virus，JEV）引起的一種蚊媒性、急性人畜共患傳染病，病豬主要表現(xiàn)為妊娠母豬流產(chǎn)、產(chǎn)死胎，公豬睪丸腫。本試劑用于檢測(cè)豬血清中豬乙腦病毒抗體，可用于豬乙腦病毒疫苗免疫效果評(píng)價(jià)。

【原 理】

 系由預(yù)包被豬乙型腦炎病毒重組E2蛋白的酶標(biāo)板、酶標(biāo)記物及其他配套試劑組成，應(yīng)用酶聯(lián)免疫法（ELISA）原理檢測(cè)豬血清、血漿樣本中豬乙型腦炎病毒抗體。實(shí)驗(yàn)時(shí)在酶標(biāo)板中加入對(duì)照血清和待檢樣本，經(jīng)溫育后若樣品中含有豬乙型腦炎病毒抗體，則將與酶標(biāo)板上抗原結(jié)合，經(jīng)洗滌除去未結(jié)合的其他成分后；再加入酶標(biāo)記物，與酶標(biāo)板上抗原抗體復(fù)合物發(fā)生特異性結(jié)合；再經(jīng)洗滌除去未結(jié)合的酶標(biāo)記物，在孔中加TMB底物液，與酶反應(yīng)形成藍(lán)色產(chǎn)物，顯色深淺與樣品中的特異性抗體含量成正相關(guān)；加入終止液終止反應(yīng)后，產(chǎn)物變?yōu)辄S色；用酶標(biāo)儀在450nm波長測(cè)定各反應(yīng)孔中的吸光值，即可知樣品是否含有豬乙型腦炎病毒抗體。

日本乙型腦炎又名流行性乙型腦炎，是由日本乙型腦炎病毒引起的一種急性人獸共患傳染病。豬主要特征為高熱、流產(chǎn)、死胎和公豬睪丸炎。

乙型腦炎是自然疫源性疫病，許多動(dòng)物感染后可成為本病的傳染源，豬的感染zui為普遍。本病主要通過蚊的叮咬進(jìn)行傳播，病毒能在蚊體內(nèi)繁殖，并可越冬，經(jīng)卵傳遞，成為次年感染動(dòng)物的來源。由于經(jīng)蚊蟲傳播，因而流行與蚊蟲的孳生及活動(dòng)有密切關(guān)系，有明顯的季節(jié)性，80%的病例發(fā)生在7、8、9三個(gè)月；豬的發(fā)病年齡與性成熟有關(guān)，大多在6月齡左右發(fā)病，其特點(diǎn)是感染率高，發(fā)病率低(20%-30%),死亡率低；新疫區(qū)發(fā)病率高，病情嚴(yán)重，

豬只感染乙腦時(shí)，臨診上幾乎沒有腦炎癥狀的病例；豬常突然發(fā)生，體溫升至40-41攝氏度，稽留熱，病豬精神萎縮，食欲減少或廢絕，糞干呈球狀，表面附著灰白色粘液；有的豬后肢呈輕度麻痹，步態(tài)不穩(wěn)，關(guān)節(jié)腫大，跛行；有的病豬視力障礙；zui后麻痹死亡。妊娠母豬突然發(fā)生流產(chǎn)，產(chǎn)出死胎、木乃伊和弱胎，母豬無明顯異常表現(xiàn)，同胎也見正產(chǎn)胎兒。公豬除有一般癥狀外，常發(fā)生一側(cè)性睪丸腫大，也有兩側(cè)性的，患病睪丸皺襞消失、發(fā)亮，有熱痛感，約經(jīng)3-5天后腫脹消退，有的睪丸變小變硬，失去配種繁殖能力。如僅一側(cè)發(fā)炎，仍有配種能力。

流產(chǎn)胎兒腦水腫，皮下血樣浸潤，肌肉似水煮樣，腹水增多；木乃伊胎兒從拇指大小到正常大小；肝、脾、腎有壞死灶；全身淋巴結(jié)出血；肺瘀血、水腫。子宮粘膜充血、出血和有粘液。胎盤水腫或見出血。公豬睪丸實(shí)質(zhì)充血、出血和小壞死灶；睪丸硬化者，體積縮小，實(shí)質(zhì)結(jié)締組織化。

由于本病隱性感染機(jī)會(huì)多，血清學(xué)反應(yīng)都會(huì)出現(xiàn)陽性，需采取雙份血清，檢查抗體上升情況，結(jié)合臨診癥狀，才有診斷價(jià)值。

無治療方法，一旦確診淘汰。做好死胎兒、胎盤及分泌物等的處理；驅(qū)滅蚊蟲，注意消滅越冬蚊；在流行地區(qū)豬場(chǎng)，在蚊蟲開始活動(dòng)前1-2個(gè)月，對(duì)4月齡以上至兩歲的公母豬，應(yīng)用乙型腦炎弱毒疫苗進(jìn)行預(yù)防注射，第二年加強(qiáng)免疫一次，免疫期可達(dá)3年。

As a result of mosquito transmission, epidemic is closely related to mosquito breeding and activities, with obvious seasonality, 80 % cases occurred in 7, 8 and 9 months. The onset age of pigs is related to sexual maturity, and most of them are around 6 months old. its characteristics are high infection rate, low incidence rate ( 20 % - 30 % ) and low mortality rate. The incidence rate of new epidemic areas is high and the disease is serious.
When pigs are infected with Japanese encephalitis, there are almost no encephalitis cases in the clinic. Pigs often happen suddenly, the body temperature rises to 40 - 41 degrees celsius, and the fever is missed, the diseased pigs' spirit shrinks, their appetite is reduced or eliminated, the dung is dry and spherical, and grayish white mucus adheres to the surface. Some pigs showed mild paralysis, unstable gait, swollen joints and limping. Some sick pigs have visual impairment; Finally paralysis and death. Pregnant sows suddenly miscarried, producing stillbirths, mummies and weak fetuses, and sows showed no obvious abnormal performance, and fetuses were also found in the same fetus. Besides general symptoms, boars often have unilateral testicular swelling, and they also have bilateral features. the scrotal folds of diseased testes disappear, shine and feel hot. after about 3 - 5 days, the swelling subsided, and some testes became smaller and stiffer, thus losing the ability of breeding and reproduction. If only one side is inflamed, there is still mating ability.
Fetal brain edema caused by abortion, subcutaneous blood sample infiltration, muscle boiling like, ascites increased; Mummy fetus from thumb size to normal size; Necrotic lesions were found in liver, spleen and kidney. Systemic lymph node hemorrhage; Pulmonary congestion and edema. Hyperemia, hemorrhage and mucus in uterine mucosa. Placental edema or hemorrhage. Congestion, hemorrhage and small necrosis of testicular parenchyma in boars; Testicular sclerosis is characterized by shrinking volume, adhesion with scrotum and substantial connective tissue.
As there are many opportunities for latent infection of this disease, serological reactions will be positive, so it is necessary to take double serum, check the rise of antibody and combine with clinical symptoms, so as to have diagnostic value.
There is no treatment method, once diagnosed, it is best to be eliminated. Do a good job in the treatment of dead fetus, placenta and secretions, etc. Eliminate mosquitoes, pay attention to eliminate overwintering mosquitoes; In pig farms in epidemic areas, sows aged over 4 months to 2 years old are immunized with attenuated Japanese encephalitis vaccine 1 - 2 months before mosquitoes start their activities, and the immunization period can reach 3 years, which is relatively high.
